It's of great challenge to address for heavy metal-contaminated soil. Once the farmland is contaminated with heavy metals, the microbial ecology of the plant rhizosphere will change, which in turn impacts crop productivity and quality. However, few studies have explored the effects of heavy metals on plant rhizosphere microbes in farmland and the role that plant cultivation plays in such a phytoremediation practice. In this study, the impacts of comfrey (Symphytum officinale L.) cultivation and the stresses of cadmium/zinc (Cd/Zn) on rhizosphere soil microflora were examined. Microbial DNA was collected from soils to evaluate the prevalence of bacteria and fungi communities in rhizosphere soils. High-throughput 16 S rRNA sequencing was used to determine the diversity of the bacterial and fungal communities. The results showed that growing comfrey on polluted soils reduced the levels of Cd and Zn from the vertical profile. Both the comfrey growth and Cd/Zn stresses affected the community of rhizosphere microorganisms (bacteria or fungi). Additionally, the analysis of PCoA and NMDS indicated that the cultivation of comfrey significantly changed the bacterial composition and structure of unpolluted soil. Comfrey cultivation in polluted and unpolluted soils did not result in much variance in the fungi's species composition, but the fungal compositions of the two-type soils were noticeably different. This work provided a better understanding of the impacts of Cd/Zn stresses and comfrey cultivation on rhizosphere microbial community, as well as new insight into phytoremediation of heavy metal-contaminated soils.

As a toxic heavy metal, cadmium (Cd) easily enters into rice while rice grains greatly contribute to the dietary Cd intake in the populations consuming rice as a staple food. The availability of Cd in paddy soil determines the accumulation of grain Cd. Soil drainage leads to the remobilization of Cd, increasing bioavailability of Cd. In contrast, soil flooding results in little contribution of soil Cd to grain Cd, which is generally attributed to sulfate reduction induced by sulfate-reducing bacteria (SRB) in paddy soils. However, effects of SRB cultured from the paddy soil on the solubility and redox behavior of Cd have been seldom investigated before. Here, we used SRB enrichment cultures to investigate the temporal dynamics of Cd2+. The results showed that SRB enrichment cultures efficiently reduced solution redox potential (Eh) to less than -100 mV and gradually increased pH to neutral, demonstrating their ability to create a good anaerobic environment. The solubility of Cd obviously decreased in the anaerobic phase and Cd2+ was transformed into poorly dissolved CdS near the SRB cell wall edge. The addition of Zn2+ and/or Fe2+ further improved the decrease in Cd solubility and facilitated the formation of polymetallic sulfides as a consequence of promoting the production of S0 and dissolved sulfides (S2-/HS-) and the transformation of S0 into S2-/HS-. Little of Cd was detected in the media upon reoxidation, which was probably due to the high pH and the interaction between CdS and ZnS/FeS. Conclusively, these results demonstrate the detailed dynamic processes that explain the essential role of SRB in regulating the redox dynamics of chalcophile heavy metals and their bioavailability in paddy soils.

Toxic metals such as lead (Pb), cadmium (Cd), mercury (Hg) and arsenic (As) that lead to many visceral organ and nervous system diseases have attracted global attention due to their gradual accumulation in human bodies. The tolerance levels of exposure to toxic metals among race/ethnic groups are different due to the variance of sociodemographic, dietary, and behavioral characteristics. Few studies focused on investigating the biomarker levels of toxic metals in different race/ethnic groups and the potential mechanisms for controlling the accumulation in human bodies. Therefore, we selected eight biomarkers for four toxic metals from the National Health and Nutrition and Examination Survey (NHANES) in the 2-year data cycle of 2015-2016 to reveal the accumulation levels in different races. According to the NHANES rules, we applied probability sampling weights. The geometric mean levels of these biomarkers were calculated in all five race/ethnic groups (Mexican American, white, black, Asian, and other Hispanic) and two Asian subgroups (U.S.-born Asian, and other-born Asian), and compared with each other. The results showed that all the biomarkers in other-born Asians were 1.1-6.7 times in blood and 1.1-3.6 times in urine higher than other race/ethnic groups. Except Hg and As, the lowest biomarker levels were recorded in U.S.-born Asians, only 0.6-0.9 times of lead and 0.3-0.8 times of cadmium than other race/ethnic groups. Furthermore, the major factors of higher Hg and As biomarker levels in Asians were dietary intake of seafood and rice, indicating different accumulation mechanisms among Asians and other race/ethnic groups, especially for U.S.-born Asians. These findings provided new insight into a deeper understanding the accumulation of toxic metals and human health.
